Gut Organoid as a New Platform to Study Alginate and Chitosan Mediated PLGA Nanoparticles for Drug Delivery

Intestinal organoids can be used as an ex vivo epithelial model to study different drug delivery effects on cells’ luminal surface. In this study, the impact of surface charge 5-ASA loaded PLGA nanoparticles into lumen was investigated. Alginate and chitosan were coat provide negative positive charges particles, respectively. The organoid growth viability not affected by presence either alginate- or chitosan-coated nanoparticles. It shown that could transported from serosal side dye gradually accumulated in day 2–3 after adding Matrigel. By 5, eliminated organoids. concluded positively charged more readily across epithelium lumen. may attributed affinity cells charge. Thus, utilized appropriate mimic functions intestinal a evaluate benefits nanoparticle-based delivery.
